Indications

Eltrombopag is utilized to address low levels of platelets in individuals diagnosed with chronic immune thrombocytopenia purpura (ITP), a specific blood disorder, or individuals with chronic hepatitis C. It may also be used to treat individuals with aplastic anemia, another blood disorder.

Platelets, which are crucial in forming blood clots and preventing bleeding, are a type of blood cell. By increasing the number of platelets, eltrombopag can reduce the risk of bleeding. Eltrombopag acts similarly to thrombopoietin, a natural substance, by stimulating the production of platelets in the body.

Uses and Dosage

Follow your doctor’s instructions when taking this medication orally, usually once a day, with or without food low in calcium. Consult your doctor or pharmacist for specific information.

Take this medication at least 2 hours before or 4 hours after consuming any products that contain magnesium, aluminum, or calcium-rich foods (containing more than 50 milligrams calcium), such as antacids, dairy products, calcium-fortified juice, vitamins/minerals, quinapril, some forms of didanosine, and sucralfate.

Follow the same guidelines for bismuth subsalicylate, iron, selenium, and zinc, as these products may interfere with the effectiveness of eltrombopag.

The manufacturer advises not to split, chew, or crush the tablet before ingesting it, and not to combine it with food or liquid. Some comparable drugs (immediate-release tablets) can be split, chewed, or crushed. Adhere to your physician’s instructions on taking this medication.

If you are using the suspension powder follow the instructions carefully when mixing and using each dose. Use the provided mixing bottle to blend each dose with cool or cold water. Gently shake the mixing bottle for at least 20 seconds to ensure the powder mixes well with water. Once mixed, use the provided special measuring device (oral syringe) to prepare and administer each dose. Do not use a household spoon, as you may not get the correct dosage. Do not reuse the oral syringe. Use a new oral syringe for each dose. If not used immediately, the mixed medication may be kept at room temperature for up to 30 minutes. After 30 minutes, discard any unused mixture or any mixture left in the bottle.

Take this medication regularly to achieve the maximum benefit. Take it at the same time every day to help you remember.

The dose is based on your medical condition, response to treatment, age, and lab tests. Your doctor will schedule blood tests (platelet counts) to determine the appropriate dosage for you. Keep all medical/lab appointments and follow your doctor’s instructions carefully.

If you are of East-Asian/Southeast-Asian descent or have liver problems, your doctor may advise starting with a lower dose because you may be more susceptible to its effects.

Stopping eltrombopag may increase your risk of bleeding. After your last dose, your doctor should order a blood test once a week for at least 4 weeks to monitor your platelet count. Notify your doctor immediately if you experience any bleeding or bruising.

Ask your doctor if you should continue taking other medications for ITP, such as corticosteroids, azathioprine, or danazol, while taking eltrombopag.

If your condition (bleeding/bruising) does not improve or worsens, inform your doctor.

Side Effects

Some side effects may occur that usually do not need medical attention:

  • back pain
  • muscle aches or spasms
  • headache
  • flu symptoms such as fever, headache, sore throat, cough, tiredness, chills, and body aches
  • weakness
  • extreme tiredness
  • decreased appetite
  • pain or swelling in the mouth or throat
  • hair loss
  • rash
  • skin color changes
  • skin tingling, itching, or burning
  • swelling of the ankles, feet, or lower legs
  • toothache (in children)

Inform your healthcare provider immediately if you experience any serious side effects:

  • swelling, pain, tenderness, warmth or redness in one leg
  • shortness of breath
  • coughing up blood
  • fast heartbeat
  • fast breathing
  • pain when breathing deeply
  • pain in the chest, arms, back, neck, jaw, or stomach, breaking out in cold sweat, lightheadedness
  • slow or difficult speech
  • sudden weakness or numbness of the face, arm or leg, sudden headache, sudden vision problems, sudden difficulty walking
  • stomach pain, nausea, vomiting, diarrhea
  • cloudy, blurry vision, or other vision changes

Caution

Inform your doctor or pharmacist about any allergies you have before taking eltrombopag, as this product may contain inactive ingredients that could cause allergic reactions or other issues.

Before using this medication, inform your doctor or pharmacist about your medical history, particularly if you have a history of blood clots or clotting problems (such as Factor V Leiden), other blood disorders (such as blood cancer, myelodysplastic syndrome), certain eye problems (cataracts), or liver disease.

Although this medication can increase platelet count and decrease the risk of bleeding, you should continue to avoid activities that could increase your risk of bleeding, such as contact sports or using sharp objects like razors and nail cutters.

If you are pregnant or plan to become pregnant, you should not use eltrombopag as it may harm the unborn baby. Please ask your doctor about reliable forms of birth control while using this medication and for seven days after stopping treatment. If you become pregnant, speak with your doctor immediately about the benefits and risks of using this medication.

It is unclear whether this medication passes into breast milk, and it is not recommended to breastfeed while using this drug due to the potential risk to the infant.
