Temozolomide is employed in the treatment of specific types of brain tumors. Classified under alkylating agents, temozolomide operates by reducing or halting the proliferation of cancer cells within your body.

Uses and Dosage

Follow your doctor’s instructions when taking this medication orally, typically once daily, according to your prescribed treatment schedule. Swallow the capsules whole with a full glass of water, equivalent to 8 ounces or 240 milliliters. Do not crush, chew, or open the capsules. In case of vomiting after taking your dose, refrain from taking any additional medication. Instead, wait and administer your next dose at the regular scheduled time.

To mitigate nausea and vomiting, take temozolomide on an empty stomach or at bedtime. For optimal efficacy, ensure consistency by taking this medication at the same time each day in relation to meals, such as one hour before or three hours after a meal.

The dosage and treatment schedule are determined based on your medical condition, height, weight, and response to treatment. Adhere to your doctor’s instructions for taking this medication. To help you keep track of your dosing schedule, consider marking your calendar.

In some cases, your doctor may also prescribe additional medications, such as antibiotics, to assist in preventing infections or managing side effects. Be sure to follow your doctor’s guidance when taking all prescribed medications.

Refrain from increasing your dose or using this drug more frequently or for a longer duration than prescribed, as doing so will not expedite the improvement of your condition and may heighten the risk of serious side effects.

In the event that the capsules are unintentionally opened or damaged, avoid inhaling the medication or allowing it to come into contact with your skin. If contact occurs, flush the affected area with water. Given that this drug can be absorbed through the skin and respiratory system and may pose a risk to an unborn baby, women who are pregnant or may become pregnant should refrain from handling this medication or inhaling the dust from the capsules.

Side Effects

Some side effects may occur that usually do not need medical attention:

• nausea
• vomiting
• diarrhea
• constipation
• loss of appetite
• sores in the mouth and throat
• headache
• pale skin
• lack of energy
• loss of balance or coordination
• fainting
• dizziness
• hair loss
• insomnia
• memory problems
• changes in vision

Inform your healthcare provider immediately if you experience any serious side effects:

• unusual bleeding or bruising
• red or black, tarry stools
• pink, red, or dark brown urine
• coughing up or vomiting blood or material that looks like coffee grounds
• fever, sore throat, ongoing cough and congestion, or other signs of infection
• unusual tiredness or weakness
• rash
• unable to move one side of the body
• shortness of breath
• seizures
• yellowing of the skin or eyes
• pain in the upper right part of the stomach
• decreased urination

Caution

Prior to initiating temozolomide treatment, inform your doctor or pharmacist if you have any allergies, whether to temozolomide, dacarbazine, or other allergens. This medication may contain inactive ingredients capable of triggering allergic reactions or causing other complications.

Before using this medication, provide your doctor or pharmacist with your medical history, especially if you have a history of kidney problems, liver problems, or bleeding/blood disorders.

Temozolomide can heighten your susceptibility to infections or exacerbate existing infections. Avoid contact with individuals carrying easily transmissible infections, such as chickenpox, COVID-19, measles, or the flu. Should you come into contact with an infection or require further details, consult your doctor.

Inform your healthcare professional that you are using temozolomide before undergoing any immunizations or vaccinations. Steer clear of contact with individuals who have recently received live vaccines, such as the flu vaccine administered through nasal inhalation.

To minimize the risk of cuts, bruises, or injuries, exercise caution when handling sharp objects like razors and nail clippers. Engaging in activities such as contact sports should also be avoided.

This medication may induce dizziness. The consumption of alcohol or marijuana (cannabis) can exacerbate dizziness. Avoid activities requiring alertness, such as driving or operating machinery, until you can safely do so.

Women and older adults may be more susceptible to the side effects of this drug, particularly an increased risk of infection and easy bruising or bleeding.

Men should refrain from donating sperm while using temozolomide and for a period of 3 months following the cessation of this medication.

Notify your doctor if you are pregnant or planning to become pregnant. It is not advisable to become pregnant while using temozolomide, as it may pose a risk to an unborn baby. A pregnancy test should be conducted by your doctor before commencing this medication. Women using this medication should inquire about reliable forms of birth control during treatment and for 6 months after the final dose. Men using this medication and whose partners are pregnant or capable of becoming pregnant should utilize condoms for birth control during treatment and for 3 months after the final dose. In the event of pregnancy, whether by the patient or the patient’s partner, promptly discuss the medication’s risks and benefits with your doctor.

The extent to which this drug passes into breast milk remains uncertain. Given the potential risk to the infant, breastfeeding is not recommended while using this drug and for at least 1 week following the conclusion of treatment. Consult your doctor before breastfeeding.
